Abstract
We have used degenerate oligonucleotide primers and PCR to clone a novel G protein-coupled receptor from rat striatum mRNA. This clone, r21, shares extensive sequence identity with the mouse delta (δ) opioid receptor identified in NG108-15 cells. Sequence analysis of r21 revealed that it is: 91%, 59%, 75% and 60% identical to the mouse δ opioid receptor in transmembrane (TM) domains III, IV, V and VI, respectively. An antisense riboprobe spanning TMs III - VI was prepared and hybridized in situ to mouse brain sections. The distribution of r21 mRNA in the mouse brain suggests that it may encode a kappa opioid receptor subtype.
