IGFs and IGF binding proteins in human breast cancer cell lines

Abstract

We demonstrated that these hBCCL have both Type 1 and 2 IGF receptors and, in data not shown, can respond to small amounts of exogenous IGFs. These cells also secrete a variety of different IGF-BPs into CM. While many of these secreted IGF-BPs appear related to the three recently characterized human IGF-BPs, the identity of the prominent 24 kD band remains unknown. It is clear that these high affinity IGF-BPs can confound attempts to detect either IGF-I or -II by radioligand assay and must be carefully removed before such assays. While it is tempting to hypothesize that these IGF-BPs act in a paracrine or autocrine manner to modulate the effects of IGF-I and -II, the physiological role of these IGF-BPs remains unclear.
