Abstract
The primary risk factor for glaucoma is elevated intraocular pressure, which results from an unidentified increase in the resistance to aqueous humor outflow through the trabecular meshwork. Glycosaminoglycans are likely contributors to the normal and glaucomatous outflow resistance. Proteoglycans (PGs), with their highly charged glycosaminoglycan side chains, are critical components of the structure and organization of the outflow resistance within the trabecular meshwork. The members of most families of PGs have been identified in the outflow pathway and their turnover and biosynthetic replacement appear to be a key component of the homeostatic mechanism of intraocular pressure regulation.
