Abstract
Purpose: We and others have reported that systemic-endotoxin-induced uveitis is accompanied by an increase in iris/ciliary body mRNA levels for several inflammation-related proteins. We have continued this project by looking at gene expression of additional cytokines, chemokines, and inflammation-related enzymes. Methods: Sprague-Dawely rats (∼ 200g) were injected in the foot pad with 200 μg endotoxin or saline. Eyes were enucleated after 3 hours for mRNA isolation and the iris-ciliary bodies were dissected for RNA extraction. Ethidium bromide staining of RT-PCR products in agarose electrophoresis gels was used to detect specific mRNAs. Constitutively expressed cyclophilin mRNA was monitored as an internal control. Results: Endotoxin induced the expression of transcripts for five chemokines (MIP-1α, MIP-10, MIP-2, MCP-1, Gro) and 2 enzymes (inducible type 2 NOS and cyclooxygenase-2). Transcripts for GM-CSF and 5-lipoxygenase were not detectable and those for cyclooxygenase-1 and 5-lipoxygenase-associated protein remained at basal levels. Conclusions: These results confirm other reports and further expand the list of genes activated in the iris/ciliary body by systemic endotoxin.
