Abstract

The R eactome Kno wledgebase ( https://reactome.org ),ãn Elixirãnd GCBR core biological data resource, provides manually curated molecular details ofã broad range of normalãnd disease-related biological processes. Processesãreãnnotatedãsãn ordered network of molecular transformations inã single consistent data model. Reactome thus functions bothãsã digitalãrchive of manually curated human biological processesãndãsã tool f or disco v ering functional relationships in data suchãs gene expression profiles or somatic mutation catalogs from tumor cells. Here w e re vie w progress to wãrdsãnnotation of the entire human proteome, tãrgetedãnnotãtion of disease-causing genetic variants of proteinsãnd of small-molecule drugs inã pathwã y conte xt,ãnd to wãrds supporting explicitãnnotation of cell-ãnd tissue-specific pathwã y s. Finally, we briefly discuss issues involved in making Reactome more fully interoperable with other related resources suchãs the Gene Ontologyãnd maintaining the resulting community resource network.
